Many great instructions are on the BasiliskII/Windows homepage:

But here's a short list of things to check in the Basilisk GUI:

-add the disk file to the installed disks (under the Disk tab)
-set your machine type to the machine you got your ROM image from
-you did get a ROM image, right? You need one. Read this:
